Fabrication of Vascularized Scaffold Containing Cardiac Tissue-derived Decellularized Extracellular Matrix Using Bioprinter
, M.Sc. Thesis Sharif University of Technology ; Mashayekhan, Shohreh (Supervisor)
Abstract
In this study, a new bioink was introduced for the production of tubular tissue structures for cardiac tissue engineering by bioprinter using the FRESH (Freeform Reversible Embedding of Suspended Hydrogels) method. The novel bioink that we used was a combination of cardiac extracellular matrix (cECM) and oxidized alginate. The cardiac extracellular matrix was used to increase the biomimetic of the printed structures to the actual tissue of the body, and also to create sites for cell adhesion, and to improve cell growth and survival. We used alginate oxide (oxidation degree: 5%) to increase the mechanical properties of the tissue. Alginate oxidation has been due to extracellular matrix (ECM)...
3D Bioprinting of Amniotic Membrane-Based Nanocomposite for Tissue Engineering Applications: Evaluation of Rheological, Mechanical and Biological Properties
, Ph.D. Dissertation Sharif University of Technology ; Simchi, Abdolreza (Supervisor) ; Tamjid, Elnaz (Supervisor) ; Niknejad, Hassan (Co-Supervisor)
Abstract
3D bioprinting is an additive manufacturing method that facilitates the deposition of the desired cells and biomaterials at any pre-defined location. This technique also enables control over the internal structure and external dimensions of printed constructs. Among various biomaterials used as bioinks, the bioinks derived from decellularized extracellular matrixes (dECMs) have attracted significant attention due to their bioactivity and being a rich source of biochemical cues. Here in this study, the decellularized amnion membrane (dAM) has been selected as the main component of the bioink formulation because of its biocompatibility, low immunogenicity, antibacterial property, abundance,...
Development of Photocured Bioinks Based on GelMa and MXene for Muscle and Neural Tissue Engineering
, Ph.D. Dissertation Sharif University of Technology ; Simchi, Abdolreza (Supervisor) ; Purjavadi, Ali (Supervisor) ; Tamjid, Elnaz (Supervisor)
Abstract
Despite the promising potential of 3D bioprinting for personalized medicine, this technology is still under development and faces challenges, such as the lack of bioinks capable of precise printing while maintaining high cell viability. The physical and chemical properties of bioinks, including mechanical, rheological, and biological characteristics, play a key role in the success of printing targeted scaffolds. In this study, photo-crosslinkable Gelatin methacrylate (GelMa)-Ti3C2 MXene (MX) bioinks were developed for muscle and nerve tissue engineering. Two-dimensional MX phase was synthesized using an in-situ HF generation method. To prepare photo-crosslinkable hydrogels, type I (IG and...
Development and Characterization of Bioinks for Cartilage Tissue Engineering
, M.Sc. Thesis Sharif University of Technology ; Mashayekhan, Shohreh (Supervisor) ; Yaghmaei, Soheyla (Supervisor)
Abstract
Meniscus regeneration remains a major challenge in orthopedics due to its structural heterogeneity and limited vascularization. In this study, a hybrid scaffold composed of silk fibroin (SF), decellularized extracellular matrix (ECM) microparticles, and polycaprolactone (PCL) was fabricated using dual-nozzl bioprinting to achieve both biological and mechanical functionality. Rheological analysis of the bioink demonstrated shear-thinning behavior, a dominant storage modulus (G′) over the loss modulus (G″), and rapid gelation confirmed by the vial inversion test, ensuring stable extrusion and shape fidelity. The bioink also exhibited structural recovery exceeding 90% after stress release....
